Overview:
Neural precursor cell expressed, developmentally down-regulated 8 (NEDD8) encodes a ubiquitin-like protein (1). NEDD8 can be conjugated to proteins via the C-terminal Gly by the E2 enzyme Ubc12 and E3 ligase complex to affect various signaling molecules such as cullins, auxin, IRF3, MDM2, and p21(2). On the contrary, the NEDD8 modified substrates can be reversed by deneddylating enzymes such as CSN, NEDP1, UAP21, Ataxin3, PfUCH54, UCHL1 and UCHL3 (3). The inhibitor of NEDD8 suppressed the tumor growth in AML cells in xenograft assays (3).
