Overview:

SENP6 is a mammalian SUMO protease that preferentially disassembles conjugated SUMO2/3 chains. It is found in the nucleoplasm and possess a conserved sequence insertion within the catalytic domain which is absent in the catalytic domains of the other SENP family members (1). SENP6 has been shown to be essential for cell proliferation and cell cycle progression with a prominent role during mitosis (2). SENP6 depleted cells exhibit defects in the spindle assembly and metaphase chromosome congression resulting in mitotic problems and cell death. Thus, SENP6 inhibitors have clinical potential against highly proliferative cancer cells (3).
