Overview:

Ubiquitin Specific Protease 1 (USP1) is a human deubiquitinase (DUB) that contains one of the largest catalytic domains within the USP family. USP1 plays a role in regulating cellular response to DNA damage and is also involved in the control of cell differentiation (1). USP1 regulates ubiquitination of Proliferating Cell Nuclear Antigen (PCNA) in response to DNA damage preventing unscheduled recruitment of error-prone TLS DNA polymerases [2]. USP1 expression is deregulated in certain types of human cancer and is thus emerging as a novel target in cancer therapy (3).
